PhD at the INRAE IJPB-Versailles: we are looking for an enthusiastic and motivated candidate that is willing to participate to a competition to get a scholarship at the doctoral school « Plant Sciences: from genes to ecosystems » (SEVE) of the Paris Saclay University. The PhD project focuses on the understanding of Arabidopsis seed metabolism and its regulation by combining genetics, gene functional analyses and metabolomic approaches. F-box proteins have diverse functions in eukaryotic organisms, including plants, mainly targeting proteins for 26S proteasomal degradation. Here, we demonstrate the role of the F-box protein SKP1-INTERACTING PARTNER 31 (SKIP31) from Arabidopsis (Arabidopsis thaliana) in regulating late seed maturation events, seed vigor, and viability through biochemical and genetic studies using skip31 mutants and different transgenic lines. We show that SKIP31 is predominantly expressed in seeds and that SKIP31 interacts with JASMONATE ZIM DOMAIN (JAZ) proteins, key repressors in jasmonate (JA) signaling, directing their ubiquitination for proteasomal degradation independently of coronatine/jasmonic acid-isoleucine (JA-Ile), in contrast to CORONATINE INSENSITIVE 1, which sends JAZs for degradation in a coronatine/JA-Ile dependent manner. Moreover, JAZ proteins interact with the transcription factor ABSCISIC ACID-INSENSITIVE 5 (ABI5) and repress its transcriptional activity, which in turn directly or indirectly represses the expression of downstream genes involved in the accumulation of LATE EMBRYOGENESIS ABUNDANT proteins, protective metabolites, storage compounds, and abscisic acid biosynthesis. However, SKIP31 targets JAZ proteins, deregulates ABI5 activity, and positively regulates seed maturation and consequently seed vigor. Furthermore, ABI5 positively influences SKIP31 expression, while JAZ proteins repress ABI5-mediated transactivation of SKIP31 and exert feedback regulation. Taken together, our findings reveal the role of the SKIP31-JAZ-ABI5 module in seed maturation and consequently, establishment of seed vigor.

Cet éclairage est apporté dans le rapport final du Groupe de travail « One Health, contributions de la santé des plantes, des sols, de l’eau, de l’air et de l’environnement » de l’Académie d’agriculture de France, daté d’avril 2023. L’Académie d’agriculture de France est, en effet, au cœur des enjeux One Health (« Une seule santé » ou « Un monde, une santé », ou encore « Une seule santé pour un seul monde »), dans une vision qui place les humains, les animaux, les plantes et les écosystèmes dans un environnement biotique et abiotique partagé. Le groupe de travail, mis en place début 2021, s’est intéressé à des aspects souvent méconnus du sujet « Une seule santé » : la santé des plantes et la prise en compte des principales composantes de notre environnement (air, eau, sol), incluant les communautés microbiennes associées. Le rapport, dont la rédaction par les académiciens signataires a été coordonnée par leur consœur Arlette Laval, apporte un éclairage riche et original sur le rôle de l’environnement dans sa diversité mais ne prétend cependant pas couvrir l’ensemble des enjeux qui lient l’agriculture à l’approche systémique de la santé. Il constitue, néanmoins, une pierre nouvelle pour la construction de cette vision systémique. Seed dormancy is an adaptive trait which enables plants to survive adverse conditions and restart growth in a season and location suitable for vegetative and reproductive growth. Control of seed dormancy is also important for crop production and food quality because it can help induce uniform germination and prevent preharvest sprouting. Rice preharvest sprouting quantitative trait locus (QTL) analysis has identified Seed dormancy 4(Sdr4) as a positive regulator of dormancy development. Here, we analyzed the loss-of-function mutant of the Arabidopsis ortholog, Sdr4 Like1 (SFL1), and found that the sfl1-1seeds showed precocious germination at mid- to late-maturation stage similar to rice sdr4 mutant, but converted to become more dormant than the wild type during maturation drying. In sync with the dormancy levels, expression levels of the seed maturation and dormancy master regulator genes, ABI3, FUS3 and DOG1 in sfl1-1 seeds were lower than in wild type at early- and mid-maturation stages, but higher at the late-maturation stage. In addition to the seed dormancy phenotype, sfl1-1 seedlings showed a growth arrest phenotype and heterochronic expression of LAFL (LEC1, ABI3, FUS3, LEC2) and DOG1 in the seedlings. These data suggest that SFL1 is a positive regulator of initiation and termination of the seed dormancy program. We also found genetic interaction between SFL1 and the SFL2, SFL3 and SFL4 paralogs of SFL1, which impacts on the timing of the phase transition from embryo maturation to seedling growth.
Depuis la Seconde Guerre mondiale, l’agronomie est devenue une discipline scientifique et technique à part entière, qui appuie le raisonnement des pratiques de production végétale. Conçue initialement comme application de la chimie agricole et de la physiologie végétale, elle a développé ses propres concepts et étendu son domaine, en intégrant les savoirs issus de nombreuses autres disciplines. Elle s’est aussi construite en étudiant les pratiques des agriculteurs et des autres acteurs de l’espace rural. Ces évolutions lui ont permis de renouveler son utilité, en répondant aux nouveaux enjeux auxquels l’agriculture n’a cessé d’être confrontée.
Comment s’est opérée cette construction ? Quels en ont été les processus, les étapes et les acteurs ? En analysant la manière dont l’agronomie s’est façonnée en France, du milieu du xxe siècle à nos jours, La Fabrique de l’agronomie vise à éclairer l’avenir et à susciter des réflexions utiles sur les conditions de son développement dans les prochaines décennies.
Cet ouvrage sur l’évolution de la discipline s’adresse notamment aux agronomes de tous métiers confrontés aux défis à venir, qu’ils soient étudiants, actifs ou retraités.
La présidente de l’université Paris-Saclay est la nouvelle ministre de l’Enseignement supérieur. C’est la première fois qu’elle fait son entrée au gouvernement.
C’est une surprise. Ce vendredi 20 mai 2022, Sylvie Retailleau, 57 ans, a été nommée ministre de l’Enseignement supérieur et de la Recherche. Depuis 2020, Sylvie Retailleau est présidente de l’Université Paris-Saclay, 13e université mondiale et meilleure française dans le dernier classement de Shanghaï paru en août 2021. C’est la première fois que cette universitaire rentre au gouvernement.

The 13th International Symposium of the French Society of Plant Biology (SFBV) aims to bring together actors interested in the biological properties of photosynthetic organisms, mainly plants, as well as their applications in agriculture and bioenergy, taking into consideration important issues such as the global climate change and the protection of the environment. At the core of this congress, there is the promotion of the plant biology field, fundamental and applied, as a whole and in its diversity. Importantly, this event aims also to establish or sustain scientific partnerships, promote exchanges between the various players in the plant biology research field and develop bridges between public research and private companies. Promoting the participation of young scientists to this type of event is also a priority of the SFBV. To this end, scholarships will be offered to students to participate to 13th International Congress of the French Society of Plant Biology and prizes will be awarded to the best oral presentations and best posters.

Présentation
Nos connaissances sur la façon dont les plantes se développent, assurent leur alimentation, fleurissent, s’adaptent à leur environnement et rendent les services que l’on attend d’elles ont progressé d’une façon impressionnante ces vingt dernières années, principalement grâce à l’essor de la biologie cellulaire, moléculaire et de la génomique. Elles laissent augurer d’autres avancées spectaculaires pour ce 21ème siècle en amélioration des plantes et gestion de l’environnement et de la biodiversité.
Nombre de ces progrès sont dus à des travaux pionniers, comme ceux entrepris par Michel Caboche (1946-2021), qui ont profondément marqué la communauté scientifique française et internationale, jetant les bases d’une véritable révolution méthodologique en biologie végétale. Pendant cette séance, en évoquant brièvement sa carrière et son impact, nous rendrons hommage à un scientifique mondialement reconnu. Puis pendant le reste de la journée les exposés scientifiques illustreront différents développements récents en biologie végétale. Ces présentations montreront comment, chez la plante modèle Arabidopsis thaliana et chez des plantes cultivées, on peut désormais mieux comprendre comment se développent les racines et les graines et sont assurées la nutrition azotée, la formation des parois cellulaires, la tolérance aux stress ou la qualité des fruits. Seront également évoquées les régulations épigénétiques et le rôle qu’elles jouent dans l’adaptation et l’évolution des plantes. Une discussion générale permettra d’aborder de nombreuses autres questions et perspectives.

Métabolites spécialisés des plantes et agriculture"

Les plantes produisent de nombreux métabolites dits « spécialisés » (MS), généralement regroupés en trois grandes catégories : les terpénoïdes, les alcaloïdes et les composés phénoliques. Certains de ces composés jouent des rôles physiologiques et écologiques majeurs tout au long du cycle de vie des plantes. Ils peuvent les protéger contre des maladies, des ravageurs et des stress abiotiques ou, au contraire, faciliter leurs interactions avec des organismes bénéfiques ou symbiotiques.
Compte tenu de ces fonctions, les métabolites spécialisés suscitent un intérêt croissant pour des applications agroécologiques allant de la protection à la nutrition des plantes, en particulier dans le contexte des changements climatiques et de développement d’une agriculture plus durable utilisant moins d’intrants de synthèse. De plus, certains de ces métabolites présentent des effets positifs sur la nutrition et la santé animales et humaines, et constituent des sources de médicaments, de colorants, d'arômes ou de parfums pour l'industrie alimentaire, cosmétique ou de la santé.
Malgré leur importance, ces composés ont été caractérisés chez un nombre limité d'espèces, et la biosynthèse, le transport ou la régulation, ainsi que les fonctions biologiques de la plupart d’entre eux restent mal connus, en particulier pour des raisons techniques (ex. quantités faibles, structures chimiques complexes). De plus, il est aujourd’hui établi que la domestication puis l’amélioration des plantes ont conduit de façon intentionnelle (ex. élimination d’impacts négatifs au niveau organoleptique ou de transformation des produits) ou non (ex. sélection pour des germinations homogènes), à une diminution drastique de la quantité ou de la variabilité de ces métabolites dans les plantes et leurs produits.
Les progrès récents des connaissances de ces métabolites et le développement des nouvelles techniques d’amélioration des plantes permettent d’envisager de réintroduire ou d’optimiser les quantités de certains de ces composés de façon raisonnée dans les plantes cultivées. Ainsi, il doit être possible de bénéficier de leurs impacts positifs en agriculture et sur la qualité des produits végétaux pour des usages alimentaires ou industriels.

Nitrogen use efficiency (NUE) is critical for the sustainable development of agricultural production. Plant NUE as an inherently complex, including N uptake, translocation, assimilation, and remobilization, is governed by multiple interacting genetic and environmental factors. High nitrogen (N) fertilization for maximizing crop yield commonly leads to postponed flowering time (heading date in rice) and ripening, thus affecting resources use efficiency and followed planting time. The seminar will introduce general views on multiple concepts and changes of NUE in temporal and space, the strategies for increasing crop NUE. The genetically controlled diversity of N fertilization effects on rice flowering time, both N uptake and physiological use efficiency, and their preliminary molecular mechanism will be presented in the presentation.

Increasing the proportion of locally produced plant protein in currently meat-rich diets could substantially reduce greenhouse gas emissions and loss of biodiversity1. However, plant protein production is hampered by the lack of a cool-season legume equivalent to soybean in agronomic value2. Faba bean (Vicia faba L.) has a high yield potential and is well suited for cultivation in temperate regions, but genomic resources are scarce. Here, we report a high-quality chromosome-scale assembly of the faba bean genome and show that it has expanded to a massive 13 Gb in size through an imbalance between the rates of amplification and elimination of retrotransposons and satellite repeats. Genes and recombination events are evenly dispersed across chromosomes and the gene space is remarkably compact considering the genome size, although with substantial copy number variation driven by tandem duplication. Demonstrating practical application of the genome sequence, we develop a targeted genotyping assay and use high-resolution genome-wide association analysis to dissect the genetic basis of seed size and hilum colour. The resources presented constitute a genomics-based breeding platform for faba bean, enabling breeders and geneticists to accelerate the improvement of sustainable protein production across the Mediterranean, subtropical and northern temperate agroecological zones. Using a high-quality chromosome-scale assembly of the faba bean genome, the genetic basis of seed size and hilum colour is explored.

Multicellular organisms undergo several developmental transitions during their life cycles. In contrast to animals, the plant germline is derived from adult somatic cells. As such, the juvenility of a plant must be reset in each generation. Previous studies have demonstrated that the decline in the levels of miR156/7 with age drives plant maturation. Here we show that the resetting of plant juvenility during each generation is mediated by de novo activation of MIR156/7 in Arabidopsis. Blocking this process leads to a shortened juvenile phase and premature flowering in the offspring. In particular, an Arabidopsis plant devoid of miR156/7 flowers even without formation of rosette leaves in long days. Mechanistically, we find that different MIR156/7 genes are reset at different developmental stages through distinct reprogramming routes. Among these genes, MIR156A, B and C are activated de novo during sexual reproduction and embryogenesis, while MIR157A and C are reset upon seed germination. This redundancy generates a robust reset mechanism that ensures accurate restoration of the juvenile phase in each plant generation. In Arabidopsis, microRNAs control the transition from juvenile to adult states. The study, using genomic, genetic and molecular approaches, investigates how miR156 and miR157 are re-activated at each generation.

Abstract: "Seed is the offspring of angiosperms. Plants produce large numbers of seeds to ensure effective reproduction and survival in varying environments. Ovule is a fundamentally important organ and as the precursor of the seed. In Arabidopsis and other plants characterized by multi-ovulate ovaries, ovule initiation determines the maximal ovule number, thus greatly affecting seed number per fruit and seed yield. Investigating the regulatory mechanism of ovule initiation has both scientific and economic significance. However, the genetic and molecular basis underlying ovule initiation remains unclear due to technological limitations. Very recently, rules governing the multiple ovules initiation from one placenta have been identified, the individual functions and crosstalk of phytohormones in regulating ovule initiation have been further characterized, and new regulators of ovule boundary are reported, therefore expanding the understanding of this field. In this review, we present an overview of current knowledge in ovule initiation and summarize the significance of ovule initiation in regulating the number of plant offspring, as well as raise insights for the future study in this field that provide potential routes for the improvement of crop yield."

Abstract: "Many over-wintering plants grown in temperate climate acquire competence to flower upon prolonged cold exposure in winter, through vernalization. In Arabidopsis thaliana, prolonged cold exposure induces the silencing of the potent floral repressor FLOWERING LOCUS C (FLC) through repressive chromatin modifications by Polycomb proteins. This repression is maintained to enable flowering after return to warmth, but is reset during seed development. Here, we show that embryonic FLC reactivation occurs in two phases: resetting of cold-induced FLC silencing during embryogenesis and further FLC activation during embryo maturation. We found that the B3 transcription factor ABSCISIC ACID-INSENSITIVE 3 (ABI3) mediates both FLC resetting in embryogenesis and further activation of FLC expression in embryo maturation. ABI3 binds to the cis-acting Cold Memory Element (CME) at FLC and recruits a scaffold protein with active chromatin modifiers to reset FLC chromatin into an active state in late embryogenesis. Moreover, in response to abscisic acid (ABA) accumulation during embryo maturation, ABI3, together with the bZIP transcription factor ABI5, binds to an ABA-responsive cis-element to further activate FLC expression to high level. Therefore, we have uncovered the molecular circuitries underlying embryonic FLC reactivation following parental vernalization, which ensures that each generation must experience winter cold prior to flowering.
